To manage (create, edit, delete etc) your profiles on how you will ship your items, follow these steps:
Click on the menu option:
Tools > Options > Shipping
A new view will open on the right hand side as shown below:

Select a site for which you wish to display your existing profiles
Select eBayUserID
Select an eBay account for which you wish to display your existing profiles

Show Profiles
Displays your existing profiles for the selected site and eBayUserID.
Buttons
Explanation
Edit selected profile
Select a profile and then click this button to edit the selected profile. The changes you make are sent to eBay and if successful, eBay will apply those changes to your listings. You don't need to edit individual listings.
Delete Selected Profiles
Lets you delete profiles no longer in use by any listing. The deletion request is sent to eBay and if successful, those profiles are deleted from eBay as well as from Wonder Lister. eBay will reject deletion if a profile is currently in use by any listing or if it is designated as the default profile. Only one profile can be designated as your default profile. Thus, you must designated another profile as your default profile before attempting to delete a profile that is currently designated as your default.
Show Active Listings
Displays all active listings that are using any of the selected profiles.
Duplicate using another eBay account
Lets you easily duplicate one or more selected profiles for another eBay account.

Shipping Details
On some eBay sites, payments can be done in multiple currencies. So, select a currency for your shipping costs.
Include / Exclude Ship to Locations
Lets you specify which locations you will or will not ship. Exclusion take priority over inclusion (in case of overlap). Domestic region can't be excluded. For details, please see below.
Domestic Shipping
Specify shipping carriers along with associated costs for shipping domestic. eBay allows a maximum of four domestic shipping carriers.
International Shipping
Specify shipping carriers along with associated costs for shipping international. eBay allows a maximum of five international shipping carriers (if Global Shipping Program option is not used) or a maximum of four carriers (if Global Shipping Program is used)

Select this option to charge specified amount irrespective of buyer's location

Select this option to charge depending upon where the buyer is located. eBay will show potential buyers a shipping calculator using which the buyers can compute the shipping cost from your location to their location.
Freight: Large items over 150 pounds
Select this option if your item is over 150 pounds in weight
First shipping service is free
Check this box if you wish to provide a free shipping service. That service must be the first one.
Use my domestic rate table
If you wish to use your rate table. The rate table must have been previously created directly on eBay's website.

Check if you wish to provide this feature. eBay requires you to meet certain preconditions if you wish to use this feature.

Check this option to use this feature. The concerned profile must have been previously created directly on eBay's website

Check this option to use this feature. The concerned profile must have been previously created directly on eBay's website
